The Core labs

The Core Laboratories of the Institute for Human Gene Therapy (IHGT) provide to its members state-of-the-art technology relevant to basic, translational, and clinical research in gene therapy. These facilities weave together apparently disparate research programs through the development, acquisition, and dissemination of critical technology.

The philosophy of the IHGT is to establish fully independent and well-funded core laboratories with dedicated staff and leadership. In addition to providing service to investigators of the Institute, the core facilities are committed to the development of improved and potentially cheaper services. Critical to their value is the continued investment in the acquisition and development of sophisticated technical services.

Cell Morphology Core

The Cell Morphology Core laboratory provides both routine and special services to support the basic research, preclinical and clinical studies performed by the members of the IHGT. The CMC staff performs routine histological services on a daily basis and has recently focused on expanding special services such as electron microscopy, immunohistochemistry and in situ hybridization. This fall the CMC staff will offer workshops on various services, microscope image capture systems and morphometric analysis.

Vector Core

The Vector Core is a critical resource to members of the Institute for Human Gene Therapy providing access to a wide array of gene transfer technology. The core has assembeled a comprehensive inventory of plasmids, cell lines, and viruses useful to the development of vectors. The Vector Core maintains strong links to key laboratories in the Institute for Human Gene Therapy involved in novel vector development. Relevant innovations are quickly brought into the vector core, where they are validated and distributed to faculty.

  • AAV Vectors
    • Large-scale prep of AAV vectors made by triple transfection, and purified by CsCl gradient
    • Large-scale prep of AAV vectors made by Ad-AAV hybrid virus, and purified by column chromatography
    • Ready-to-use AAV vectors carrying reporter genes (LacZ, GFP, AFP, and A1AT)
    • QC assays on AAV vectors: genome titer and IC assay

    • Large-scale prep of plasmid DNA
