There are now two sections to PNACL's DNA Sequencing Trouble Shooting Guide!

Basically this is a compilation of all the problems we see in our DNA Sequencing Facility here at the University of Leicester. This section will continue to expand as we see more problems, which are never ending in their variety.

Note. All images are simple PICT files taken from Sequencing Analysis v3.0 which have then been converted to GIF format. To see the images up-close just click and hold on the image, then open it up. All data was produced on Graeme - our ABI 377XL DNA Sequencer using BigDye Terminators this may lead to some differences in patterns associated with different dye sets (Rhodamine and dRhodamine), although most fixes given here will work for all dye types. Please consult your PE Biosystems tech support office for details! Please also note that if you are not using the Sequencing Analysis software you will not be able to see any raw data, only the analysed version.

Analysed Data. If you are having problems with your sequencing then this section is for you! We've put examples of some quite common mistakes in this section, and tips on solving them too.

And of course there is still the famous 'Gel Problems' page. This is more for users of the 377 sequencers themselves, but we intend to add more useful information for users of our service in the near future

Just to point out that this is in no way any sort of official trouble shooting guide for PE-Biosystems 377(XL) instruments. All solutions given here are probably one of many ! If they don't work for you, then try the ABRF for more information. In fact, become a member it's an excellent resource.
