Silver Staining of SSCP gel

(according to a protocol from Lilianna Solnica-Krezel Lab)

0. Prepare 500 ml of each solution
1. Remove the larger glass plate with a spatula.
2. Place a sheet of gel support film (ATTO, AE-3355) with the adhesive side onto the gel, Make a firm contact with a roller (such as a disposable pipet).
2. Cut the side of the gel to use a minimal area for staining.
- Fix the gel with 10 % acetic acid for 20 min.
- H2O 2 min x 3 times
- AgNO3 (1g/L), 1.5 ml 37% HCOH / L (formaldehyde), for 30 min
- H2O 20 sec (optional)
- Na2CO3 (30 g/L), 1.5 ml HCOH / L (formaldehyde), Na2S2O3 5H2O (2 mg/L)*, for 2-5 min
* Weigh 20-100 mg, add H2O at 20-100 mg/ml, use 1mg eq. for 500 ml of solution
- stop with H2O wash 2-3 times, and fix with 2% AcOH, for 15 min, wash with H20
- Dry the gel at 80 oC for 1hr
