PROTOCOL FOR DILUTION OF PYROCIDEŽ FOR THE BOTTLE BIOASSAY

PHEREC Technical Memorandum # 13

Start with the label.

The manufacturer's label states that PyrocideŽ contains 5% pyrethrin.  That is, 5 parts per hundred of AI.

1.  First, we will prepare a STOCK solution.  Take two (2) ml of "off the shelf" PyrocideŽ and place into a 100 ml volumetric flask.  Add 98 ml of A.C.S. acetone to make 100 ml final volume.  This is the STOCK solution.  It contains 1000 parts per million [ppm] pyrethrin.

2.  Next, we will prepare the test solution.  Take three (3) ml of the STOCK solution and place into a 100 ml volumetric flask.  Add 97 ml of A.C.S. acetone to make 100 ml final volume.  This is the TEST  solution.   This quantity is equivalent to 30 ppm.  30 ppm is the diagnostic dose based on recent range finding tests conducted at Florida A&M University.

3.  Use one (1) ml of the TEST solution per Wheaton bottle for the bottle bioassay.  Follow the standard protocol provided.

4.  The calculations given in this protocol are preliminary and are subject to modification based on further testing.  This is a DRAFT PROTOCOL.  Please check back for updates.

This protocol was drafted as part of a two-day workshop conducted May 22-23, 2007 at Benton County Mosquito Control, West Richland, Washington.  Kevin Shoemaker organized the workshop which was generously supported by Clarke Mosquito Control and Valent Biosciences.   Particular thanks go to Bob Boggs and Stephanie Whitman.
