The director referenced the film's contagious theme song in the caption of the image, which read: '#whoyougonnacall.'
Posing beside the famous car, Melissa crossed her arms while Leslie stood beside her, her hands gripping her proton pack.
On the other side of the film's iconic logo, Kate slung her hands around the anti-ghost weapon while standing next to Kristen.
Spirit slaying machine: Paul took to his Twitter on Tuesday night to show off the newest edition to the Ghostbusters reboot cast - the Ecto-1 car
Melissa also shared Paul's tweet, including her own caption '#Ghostbusters #WomanPower.'
The photo is just one of a few images Paul has been releasing from the Ghostbusters set in recent days.
On Tuesday night, the director took to Twitter to show off the newest edition to the Ghostbusters reboot cast - the Ecto-1 car.
The OG: The original franchises' busters used a 1959 Cadillac Professional, complete with tail fins, the new all-female crew have got an update
Posting multiple pictures of the car, the Hollywood heavyweight captioned the snaps '#whattheygonnadrive'.
The new version of the Ecto-1 has retained the same red and white colour scheme and the classic Ghostbusters' logo.
But while the original franchises' busters used a 1959 Cadillac Professional, complete with tail fins, the new all-female crew have got an update.
Creating buzz: Posting multiple pictures of the car, the Hollywood heavyweight (pictured April) captioned the snaps '#whattheygonnadrive'
However, while the vintage car may have been replaced with a newer model, the Ecto-1 is hardly a car from the future.
Instead the team will be heading to ghost emergencies in what appears to be a Cadillac from the Eighties.
The boxy shape of the car seems to be that of a Cadillac SS Victoria - the go to model for hearses.
No streams crossed: The director also revealed the Proton Packs on his social media last month
Ghostbusters suits: The 52-year-old filmmaker previously teased fans by tweeting a photo of the costumes worn by his all-female cast writing, '#Whatyougonnawear'
The big car reveal came as it was announced that Ady García and Michael K. Williams officially joined the cast.
Filming is already underway on the new movie, with the female-dominated cast seen in the flesh, for the first time last month as they acted out scenes on the Boston, Massachusetts set.
Kristen was wearing seen wearing a wig to play her character Erin Gabler, who will not be seen on screen until next year.
Ready for action: Filming has already started on the reboot and Kristen is seen here on June 30 on the Boston, Massachusetts set
Melissa and Kate, who play Abby Bergman and Jillian Holtzmann respectively, hammed it up as they shot a scene in which they were either fleeing or chasing their antagonist.
Leslie meanwhile plays Patty Tolan while Chris Hemsworth is playing the ghostbusters' secretary, a role that Annie Potts filled in the 1984 original.
Dan Aykroyd, who starred in the original Ghostbusters, recently revealed that he's read the script for the sci-fi reboot and told Extra: 'Oh man, it's hot. I read the script and those girls are gonna kill it.'
Dan's original Ghostbusters was budgeted at $32m and has grossed more than $295m worldwide
He also has a writing credit on the new film.
