They play the best of friends, living life through thick and thin in their latest film.

And Miss You Already co-stars Drew Barrymore and Toni Collette could not wait to reunite off the set, as the pair demonstrated at the premiere of their film at the Toronto International Film Festival on Saturday.

The 40-year-old actress beamed in delight when she approached Toni, 42, before pulling her in for a warm and tight embrace.

Drew and Toni looked equally fantastic in their mismatching garments.

True to her free-spirited form, Drew donned an exotic blue floral dress that had a cut-out along her low back and skimmed the red carpet with a modest, sheer train.

Her sandy blonde locks tumbled down in its usual wild and beachy hairdo, and her youthful complexion glowed with a rosy hue of blush and a flattering slick of jet black eyeliner.

Toni was Drew's polar opposite with her chic mustard yellow skirt that draped on the floor and flattered her slender waistline.

The sun-kissed Australian actress swept her cropped pixie haircut across her forehead in a slightly edgy 'do, and she highlighted her healthy tan with bronze eye shadow and a matching shade of blush. 

Large and ornate silver earrings glammed up her complexion, and she carried a marble inspired clutch.

Once Drew and Toni joined Miss You Already director Catherine Hardwicke, the two stars erupted into laughter. 

The co-stars fiercely squeezed each other once again, and Catherine stood beside the pair, smiling and appearing to clap.

In Miss You Already, Drew and Toni portray childhood friends, who remain attached at the hip even in their adult years.

However, Toni's free-spirited character Milly is abruptly faced with a cancer diagnosis, while Drew's character Jess finally finds herself pregnant after various attempts. 

During an interview with Entertainment Tonight Canada at the premiere, Drew revealed she identified with the role of Milly.

'I was channelling all my friends who are much more of like, the mature rocks in my life, and I feel like I've always been almost more like Toni's character, just more, I don't know, tornado-ish,' Drew told the website. 

'I really channelled the people that I've always looked up to and wanted to become like. And hopefully I am more like that person in my life now and so I pretended to be that person in this film,' she added. 

Miss You Already will hit U.S. cinemas on November 6.
