Think 'make them happy, make them happy, make them happy' and you're on the right track! The best navigation design for your ecommerce website should be this: it's either all on top or all on the left. It has been shown that people naturally look for navigation links at the left-hand side of the web site and if they don't find any, they look for it at the top.
If they don't, they will look for navigation at the bottom. Take note that hardly anyone ever look for navigation links on the right side of the page. If you put it there, it's easy for your potential customers to miss it and convince themselves that you've got absolutely nothing in there.

'Buy Now' buttons simply refers to a button that people put on their web sites to encourage people to buy - lure them to click on the button only to be taken to the shopping cart. It shows from research that using an attractive looking 'Buy Now' button for your ecommerce web site is better than simply using a text link that says, 'Buy here'.
Make it attractive enough to entice them to click on the button, that should be enough.
